Comparison of Growths of Fastidious Bacteria in Columbia Broth, TSB and BACTEC 16A Medium




Abstract
Blood culture media should provide sufficient nutrients and suitable environment for the rapid growth of various bacteria . In this study, Columbia broth(CB), soy bean casein digest medium(Tryptic soy Broth, TSB) and BACTEC 16A medium were
compared
for
their capabilities to support growth of fastidious bacteria. Ten-fold dilutions of actively growing organisms were inoculated into 30-ml broth and incubated at 35¡É. Macroscopic growth of Streptococcus pyogenes , S. agalactiae, S. pneumoniae, L.
monocytogenes and Haemophilus aphrophilus were relatively faster with all of the 3 media, but growth value(GV) by BACTEC NR-660 system were higher with 16A. Growths of Campylobacter fetus subsp. Fetus , Cardiobacterium hominis, DF3, H.
actinomycetemcomitans, H. influenzae, Kingella denitrificans and P. multocida were slow, but relatively higher GVs with 16A and slightly more rapid macroscopic growth with CB were noted. It is concluded that better results may be obtained with CB
in the
conventional blood culture and with 16A in the BACTEC method
